duke.eduStemcellsundergodynamicchangesinresponsetoinjurytoregeneratelostcells.However,theidentityoftransitionalstatesandthemechanismsthatdrivetheirtrajectoriesremainunderstudied.Usinglungorganoids,multipleinvivorepairmodels,single-celltranscriptomicsandlineagetracing,wefindthatalveolartype-2epithelialcellsundergoingdifferentiationintotype-1cellsacquirepre-alveolartype-1transitionalcellstate(PATS)enroutetoterminalmaturation.Transitionalcellsundergoextensivestretchingduringdifferentiation,makingthemvulnerabletoDNAdamage.CellsinthePATSshowanenrichmentofTP53,TGFβ,DNA-damage-responsesignallingandcellularsenescence.GainandlossoffunctionaswellasgenomicbindingassaysrevealedadirecttranscriptionalcontrolofPATSbyTP53signalling.Notably,accumulationofPATS-likecellsinhumanfibroticlungswasobserved,suggestingpersistenceofthetransitionalstateinfibrosis.Ourstudythusimplicatesatransientstateassociatedwithsenescenceinnormalepithelialtissuerepairanditsabnormalpersistenceindiseaseconditions.
